Anticancer Activity of Ontario Grown Onion Varieties: An In-vitro Study
Description: Dietary flavonoids have played an important role in promoting the health and wellbeing of individuals and have the potential to lower risk of chronic illnesses such as diabetes and cardiovascular diseases. Onions are a major dietary source of flavonoids. However, due to their genetic diversity and differences in environmental conditions, screening for the health benefits of phenolic and flavonoid content specifically those of Ontario’s grown varieties are required. In this study, we examined the cytostatic capacity of five Ontario grown onion varieties namely Stanley, Safrane, Fortress, Lasalle and Ruby Ring on a human colorectal adenocarcinoma cell line, Caco-2. Colorimetric assays, LDH and MTS tetrazolium measured the cytotoxic and anti-proliferative effects of Ontario grown onion varieties’ extracts on Caco-2, respectively. End points were evaluated for various extract exposure times and were compared to a commercially available flavonoid. Onion extracts demonstrated varying levels of cytostatic effects on Caco-2 proliferation. Differences in cellular morphology and fluorescence intensities of key intracellular organelles between treated and untreated Caco-2 cells were imaged using fluorescence microscopy. These provided a better insight on the extracts’ possible mode of action. Identifying the best onion varieties and boosting their anti-proliferative potency by optimizing their growth, storage, and extraction will showcase onion extracts as high value chemicals. This will enhance their marketability as possible anticancer agents in cancer treatment regimens or as nutraceutical products to protect individuals against the onset of cancer.
